Description:
Methisoprinol is a chemical complex formed from inosine and an aminoalcohol, dimethylaminoisopropanol at a 1 : 3 ratio. It is a white powder, slightly bitter, water soluble and stable in a neutral solution. This complex was discovered by Gordon, Glasky, Lynes and Doty, American researchers. lsoprinosine® increases protein and nucleoprotein synthesis; preserves the structure and function of the polyribosomes; inhibits ribonucleases; reinforces polyribosome function; interferes with the genetic transfer of the virus to the polyribosome of the host cell and in this way stops viral multiplication. From the clinical studies made by Argentine researchers emerges the non-specific antiviral action of wide spectrum (Ink, Antonini, Andres, Stefano and others).
Indication:
Viral diseases of the respiratory tract, exanthemic diseases, liver diseases and some nervous diseases. Pediatric grippe, adult influenza, common cold, bronchiolitis, rhinopharyngitis, Varicella, measles, herpetic of Aphthous stomatitis, hepatitis, parotitis, etc.
Dosage and administration:
Under five years old, and average of 50 mg/ kg body weight/ day. This dosage can be increased up to 100 mg/ kg body weight/ day depending on the gravity of the case. Above five years, old and adults, an average of 50 mg/kg body weight/ day will be used, this dosage can be increased up to 60 mg/ kg body weight/ day. These dosages should be divided into four to six times a day. Should it be necessary, due to microbial complication, to use antibiotic or chemoterapeutics there are no contraindications and it will be left to the doctor's judgement. Treatment duration will be as follows.
- Acute viral diseases of short evolution - treatment should and continue one to two days after the symptomatology disappears, according to the doctor's judgement. In this way, relapses will be avoided.
- Acute viral diseases of long evolution - treatment should continue one to two weeks after the symptomatology disappears, according to the doctor's judgement. In this way, relapses will be avoided.
Care should be taken when administering Isoprinosine® (methisoprinol) to gout patients, since it increases serum uric acid contents slightly. Since it has an evident slight cardiac effect, care should be taken when administering to cardiac patients under digitalis. It should not be used on microbial infection diseases since it does not have antibacterial action. Since the ratio between therapeutic and toxic dosages is 1 : 100, Isoprinosine® can be used without fear of toxic action. Accumulation does not occur since it is rapidly eliminated together with its metabolites through the urine. It should not be used during the first four months of pregnancy. ;

Uraian:
Methisoprinol adalah suatu kompleks senyawa kimia yang terbentuk dari inosine dan suatu aminoalcohol, dimetilaminoisopropanol, dengan rasio 1 : 3. Isoprinosine® berupa bubuk putih, sedikit pahit, larut dalam air dan stabil dalam larutan netral. Kompleks ini ditemukan oleh para peneliti dari Amerika: Gordon, Glasky, Lines dan Doty, Isoprinosine meningkatkan sintesa protein dan nukleoprotein; melindungi struktur dan fungsi poliribosom; mendiami ribonuklease; menguatkan fungsi poliribosom; menghalangi pemindahan genetika virus ke poliribosom sel tubuh dan.dengan cara ini menghentikan multiplikasi virus. Dari studi klinis-studi klinis yang dilakukan oleh para peneliti Argentina, didapatkan aktivitas anti virus non spesifik dan berspektrum luas (Ink, Antonini, Andres, Stefano dan lain-lain).
Indikasi:
- Infeksi virus pada saluran napas, penyakit-penyakit eksantem.
- Penyakit hati, dan beberapa infeksi sistem saraf.
- Influensa pada anak-anak dan orang dewasa.
- Common cold.
- Bronkiolitis.
- Rinofaringitis.
- Varisela.
- Campak.
- Herpes simplex dan herpes zoster.
- Parotitis.
Posologi:
Usia di bawah lima tahun rata-rata 50 mg/ Kg BB/ hari. Dosis ini bisa ditingkatkan sampai 100 mg/ Kg BB/ hari, tergantung pada beratnya infeksi.
Usia lima tahun ke atas dan dewasa: rata-rata 50 mg/Kg BB/ hari, yang dapat ditingkatkan sampai 60 mg/Kg BB/ hari. Semua pemakaian dalam dosis terbagi 4 - 6 kali sehari. Jika perlu, antibiotika atau khemoterapi dapat digunakan bersama-bila ada infeksi bakteri. Tak ada kontraindikasi pemakaian bersama antibiotika/ khemoterapi dan ini tergantung pada penilaian dokter. Lama pengobatan sebagai berikut:
- Infeksi virus akut yang berlangsung singkat : pengobatan harus dilanjutkan satu sampai dua hari setelah gejala-gejala mereda, sesuai dengan penilaian dokter.
- Infeksi virus akut yang berlangsung lama: pengobatan hendaknya dilanjutkan satu sampai dua minggu setelah gejala-gejala menghilang, sesuai dengan penilaian dokter. Dengan cara ini kekambuhan akan dihindari.
Hati-hati ketika memberikan Isoprinosine®
melalui urin.
Isoprinosine® hendaknya tidak digunakan selama empat bulan pertama kehamilan.
